Solar powered robotic sunfish grooves to the Grateful Dead

5 hrs.

A proof-of-concept solar-powered robot modeled after the giant ocean sunfish could one day serve as biological sensor capable of keeping tabs on the environment.

A video released this week by the robot?s maker, senior scientist Tom Zambrano at AeroVironment, shows it swimming in Pacific Ocean as it grooves to the Grateful Dead?s Bird Song.

The delightfully chill viewing experience provides a few details about the robot, called Mola, which is derived from the scientific name of the giant ocean sunfish, Mola mola.

The fish are the largest known bony fish. They will sometimes float to the ocean surface and bask on their long, flat sides. That behavior appears to be the inspiration for the robot.

Mola looks as if its little more than flat solar panel with fins. As shown in the video, solar panels can be attached to rear to make a flexible tail that supplies additional power.

The panels collect enough power from the sun to cruise through the water at 2 knots as a data logger records physical, chemical, and biological information.

We?ve reached out to Zambrano for additional information on the robot and will update if we learn more.?

For now, watch the video and enjoy the high notes from the Grateful Dead. Given Jerry Garcia?s fondness for scuba in his later years, the song selection for the video clip feels alright.

What might have been in Eve?s report on WWE?s tag team division?

As SmackDown General Manager Booker T handled the fallout of the tag team free-for-all that transpired on the Aug. 24 edition of SmackDown (WATCH), Eve made quite a first impression as the five-time WCW Champion?s new assistant. With Booker asking for recommendations as to who should be the new No. 1 contenders to face WWE Tag Team Champions Kofi Kingston & R-Truth, the assertive Diva produced a research binder including each possible challenger?s recent win-loss record ? as well as her personal recommendations, which noticeably impressed the leader of the blue brand. (WATCH)

So who did Eve include in her report? How did she summarize the candidacy of each team as they chase Kofi Kingston & R-Truth for their championship gold? What other information did she include to ensure each potential contender receives fair consideration from the SmackDown GM?

While the results of her study were meant for Booker?s eyes only, the pairings Eve included and the criteria she deemed important remain open to speculation. As such, WWE.com dug into its own scouting reports to deduce how the former two-time Divas Champion likely classified the contenders and pretenders in WWE?s tag team ranks.

Flake wins Arizona GOP nomination for US Senate

PHOENIX (AP) ? With the easy defeat of an unexpectedly aggressive primary challenger, Arizona Rep. and GOP Senate nominee Jeff Flake brushed off speculation of a bruised candidacy with a vow to repeal President Barack Obama's economic and health care policies.

"This will be a great race because there are big differences between our philosophies," Flake said of Democratic opponent Richard Carmona, a moderate who served as surgeon general under Republican President George W. Bush. "... Richard Carmona is President Obama's hand-picked candidate for Arizona, and he shares the president's agenda. And I don't think it is the agenda Arizonans want."

Flake took 70 percent of the Republican vote Tuesday, defeating wealthy businessman Wil Cardon and two lesser-known candidates for the nomination to succeed retiring Republican Sen. Jon Kyl.

Democrats are hoping the accusations of hypocrisy, broken promises and flip-flopping that marked the GOP primary weakened Flake and his bank account enough to give Carmona the best shot in years at taking one of the state's two Senate seats. Arizona hasn't elected a Democrat to the Senate since Dennis DeConcini won his third term in 1988.

The push by Carmona comes as the GOP is fighting to pick up four more seats to wrest control of the chamber in advance of votes on key policy issues such as the possible repeal of President Barack Obama's health care law and changes to the tax code.

Flake said he planned to focus on the economy and the repeal of Obama's health care plan.

One or both major parties also had primary races in all nine of Arizona's U.S. House districts, including a two-incumbent rivalry between freshmen Republicans pitted against each other by redistricting. Rep. David Schweikert defeated former Vice President Dan Quayle's son, Rep. Ben Quayle.

And Rep. Ron Barber, a former aide who was hand-picked to succeed Gabrielle Giffords after she was shot in the head in a January 2011 mass shooting in Tucson, fended off a challenge by former state Rep. Matt Heinz in the Democratic primary.

Primary voters in Alaska, Oklahoma and Vermont also decided House races.

In Arizona's Senate primary, Cardon tried to paint Flake as a Washington insider who reneged on past promises to limit his terms in office. Opponents pointed to Flake's little-known past as a Washington lobbyist for a uranium mine that was minority-owned by Iran. He also criticized Flake, saying he made a dramatic change on immigration, going from a supporter of comprehensive reform to backing policies that would first secure the border.

For his part, Cardon was accused of painting himself as tough on immigration, while a company he partly owned was fined for faulty paperwork in hiring workers with questionable legal status.

Cardon, who spent $6 million of his own money, said he believed Flake was "a better candidate because of the fights he had to go through."

"We had a good campaign," Cardon said. "Unfortunately we didn't get through to all the voters."

In Oklahoma, Westville plumbing company owner Markwayne Mullin defeated three-term state Rep. George Faught in a runoff for the Republican nomination for the seat being vacated by Democratic Rep. Dan Boren. And longtime state and federal prosecutor Rob Wallace, of Fort Gibson, beat Muskogee seed company owner Wayne Herriman in the Democratic runoff.

Windsor businessman John MacGovern handily defeated a Republican primary opponent in the Vermont race to challenge independent U.S. Sen. Bernie Sanders in November.

In Alaska, state Rep. Sharon Cissna of Anchorage was leading a five candidate field of Democrats vying for the chance to challenge Republican U.S. Rep. Don Young, who has held his seat for 40 years.

Las Vegas burlesque star Holly Madison pregnant

LAS VEGAS (AP) ? Burlesque performer and Las Vegas Strip star Holly Madison says she's going to be a first-time mom.

The 33-year-old star of "Peepshow" told Us Weekly she and her boyfriend of nine months, Electric Daisy Carnival rave founder Pasquale Rotella, are expecting. Rotella issued a statement saying the two are in love and counting the days until the birth.

Madison previously announced that she will finish her run with "Peepshow" at the Planet Hollywood Resort and Casino on Dec. 30. Show officials say tickets are still available through the end of the year, but they're working with Madison to determine how much of that time she'll be performing. She's 12 weeks into her pregnancy.

"Peepshow" debuted in March 2009.

Madison previously worked for Playboy and dated the magazine's creator, Hugh Hefner.

Golf Lessons For Beginners - Getting Your Drives On The Fairways

If you can learn to master your golf shots so that you hit the ball accurately you will find that your statistics for hitting the fairways and greens in regulation will dramatically increase. This in turn translates into playing better golf all round. Here are some tips I found from reading golf lessons for beginners from the top players who have game plans to keep the ball on the short grass. Although it is exciting to watch some top players scramble from the most horrendous of situations, as an amateur, it is better to avoid that stress, and discover ways in which you can find more fairways.

As you can hit the ball further with your driver there is potentially a greater chance of the ball going astray. This means that it is harder to hit your target. Think about the wind direction and how that will impact on the ball direction. Find out where there may be traps which you need to avoid. Then choose a spot on the fairway as your target. Visualize your ball flying in the air so that you see the shot before you play it. Remember to keep your golf swing the same for the driver as you would for your irons. The power in your driver swing comes from the longer swing arc so don't try to hit it harder.

When driving distribute your weight equally over both feet as you address the ball. An error at set up can lead to other faults. Beginner golf lessons always emphasize the concept of sticking with sound fundamentals. Make sure that most of your weight is not resting on your heels as this would then mean you are too far away from the ball. This would then affect the path of your club as you take it away and your weight shift will not be correct with the result of a poor shot - either a low cut or pulling the ball. The tip here sounds simple but many players have too much weight either on their heels or balls of the feet.

At the top of your backswing try to achieve a well wound up position. Your weight should be focussed on your right side. The weight is then shifted to the left side as you make your downswing. Keep the club out in front of you as make the downswing. If it gets stuck behind your body on the downswing then your shot will not be a good one.

So, there are a couple of tips that I found from some beginner golf lessons to help you find more fairways and avoid having to make those scrambling shots.

As gamers flock to 'Guild Wars 2,' some get the boot

8 hrs.

The highly anticipated online fantasy game "Guild Wars 2" officially opened its doors today and already hundreds of thousands of players have flocked to the game.

Not surprisingly, gamers?and the game's developers at AreneaNet have hit a few bumps along the way?as hordes of players?have tried to sign in?to the massively multiplayer online role-playing game.?

Some players are?reporting problems connecting to the game while others are having issues creating characters in?the game, which takes place in the virtual, magic-filled?world of Tyria.?Meanwhile the developers at ArenaNet say they've already?had to suspend some "Guild Wars 2"?players for offensive and hateful behavior.

As they launched the game Tuesday,?ArenaNet officials announced that one million people?had already?played "Guild Wars 2" from Saturday through Monday?during a?three-day headstart program that launched over the weekend. Those who pre-ordered "Guild Wars 2"?were allowed to start on?Saturday. And ArenaNet reps say?that,?at one point,? 400,000 people were playing the game at one time.?

In fact, one French player?managed to get his character to the game's top level (level 80) in a record-breaking 32 hours of playtime.

But some players?on Tuesday reported having trouble logging into the game.?The official @GuildWars2 Twitter account acknowledged the issues?tweeting: "We're aware of the login issues and are working on them???thanks for your patience!"

ArenaNet also assured European?players that they are aware of connectivity issues there and tweeted, "We are currently pushing updates to fix bugs or issues that are affecting gameplay." And in the afternoon they?added, "For those of you having problems with lag or character creation, we're looking into it. Thanks for letting us know!"??

Meanwhile,?players have reported issues with the "Guild Wars 2" overflow server system, which is?designed to allow people?to keep playing when their own server is overloaded. And the "Guild Wars 2" Trading Post ?-- a system that allows players to trade items between?each other -- is offline.

ArenaNet issued the following explanation:

We currently have a couple serious bugs with one of our important back end systems. This error is causing the loss of trading post, players to sometimes be separated and unable to travel to each other when switching maps and entering overflow servers, and numerous guild errors. We are very sorry for this inconvenience and are currently working around the clock to try and remedy this error.

When this bug is fixed overflow servers will keep parties together and players will be able to travel to the same overflow server as any friend or guild member who is in an overflow.

Also offline: The official "Guild Wars 2" forums. When asked about it,?ArenaNet tweeted that, "at present our main focus is getting players in game."

With hundreds of thousands of players trying to jump into?a brand new online?game, these kinds of issues?are not out of the ordinary. When "Diablo 3" arrived?earlier this year, Blizzard ended up?apologizing to the many?gamers?who?encountered?servers so?overwhelmed by the launch-day crush the game was unplayable. And when BioWare launched "Star Wars: The Old Republic" servers quickly became over?crowded?and?many?players found themselves stuck in waiting rooms.

But the kinks and bugs associated with a new online game aren't the only issues facing the?"Guild Wars 2" creators and players.

Some players have found themselves suspended from the game for giving their characters offensive names and for using offensive and racist language while chatting in the game. It has become such a hotly discussed topic, the folks at ArenaNet took to Reddit to explain why they have?banned players?and to?offer examples of exactly the kind of names and language that can't be used.

They wrote:

To keep Guild Wars 2 a pleasant place to be, we take action against racist names, hate speech, and other unacceptable behavior. We have suspended some accounts involved in the use of offensive character names or inappropriate chat. The number of account blocks is miniscule: less than .001% of our total player base.

When an account is blocked for a chat offense, the account is given a three-day suspension. When an account is blocked for an offensive name, the player is required to rename the character name and, in most cases, the account is also given a three-day suspension.

In the lively Reddit discussion, ArenaNet went through some of the names and comments that earned a ban. Most are not appropriate for me to print here, but,?for example,?ArneaNet officials explained?it's not ok to say the following:

  • "Oh I am gonna break dance on your anal intercorse" (sic)
  • "shut the f(bleep) up with non-english, geez"
  • "the worm IS f(bleep)ing hard if you're a f(bleep)ing mentalpatient no we f(bleep)ing dont you can take a keep with 5 people if you're not a f(bleep)ing?(bleep)head"

Also, ArenaNet officials advise, "Save yourself a possible headache and avoid real-world and celebrity names." And no, you can not use the names?"Stop It Daddy Ahhhh" or "Adolf Critler."?In case you were wondering.

Though the game has now launched, the folks at ArenaNet say their work has only just begun.

As Mike O'Brien, president and co-founder of ArenaNet, said in a statement: "Now our focus is shifting to building on this living world we've delivered, continuously refining our game and improving services to ensure our players have the best possible experience."?

Winda Benedetti?writes?about video?games for NBC?News. You can follow her tweets about games and other things?on Twitter?here?@WindaBenedetti?and you can?follow her?on?Google+.?Meanwhile, be sure to check?out the?IN-GAME?FACEBOOK PAGE?to discuss the day's?gaming news and reviews.

Doing the math behind homeschooling ? Schools of Thought - CNN ...

by Carl Azuz, CNN

(CNN) -?Public, private, parochial, charter schools: There's no shortage of options on where to send your children for their education.

But a growing number of Americans are choosing not to send them anywhere at all, opting instead to educate them at home.

The National Center for Education Statistics says that 1.7 percent of kids were homeschooled in 1999, 2.2 percent in 2003, and 2.9 percent in 2007. Today, that figure is at 4 percent, according to an article published at EducationNews.org.

So it appears that the homeschooling growth rate is more exponential than it is steady.

Most parents aren't certified teachers, so it stands to reason why some question the effectiveness of a homeschool education. But the Home School Legal Defense Association, an advocacy group in favor of homeschooling, reported in 2009 that homeschooled students averaged 37 percentile points higher on standardized tests than their public school counterparts.

EducationNews.org backs that up, saying that while students in traditional schools mark the 50th percentile on standardized tests, students who are ?independently educated? score between the 65th and 89th percentile.

Of course, there?s a time commitment involved in homeschooling that many families simply can?t make. If a single parent has a full-time job ? or if both parents do ? setting aside several hours a day to educate a child simply isn?t feasible.

And the arguments against homeschooling ? from varying state requirements to reduced social interaction among peers to a lack of student competition ? can be challenging issues to address.

But if the number of kids who are homeschooled continues to rise, it may signal a noteworthy trend.

Pro-Palestinian activists barred from West Bank visit

Israel and Jordan on Sunday barred pro-Palestinian US and European activists from trying to cross into the West Bank to deliver aid to students, a number of sources said.

Olivia Zemor, spokeswoman for the "Welcome to Palestine" group spearheading the mission, told AFP that one bus of activists was allowed through the Allenby Bridge border crossing by the Jordanian authorities but the Israelis turned them back.

"The (Israeli) authorities took all their passports and stamped 'entry denied' on all of them and told them to return," she said.

"The second bus was prevented (from crossing) by the Jordanians at the request of the Israeli authorities. They obeyed the orders of the Israelis," she said.

"One police officer told us that 'we got orders from Israel. We cannot let you leave'. We demanded explanation and nobody gave us any explanation. Then some of us demonstrated," she said.

An Israeli defence ministry official in Jerusalem confirmed that Israeli authorities had stopped the activists from entering the West Bank after a group of them arrived at the Allenby Bridge crossing.

"A small group of provocateurs, rioters and known troublemakers arrived at the Allenby crossing earlier today," the official said.

"The state of Israel exercised its right to refuse their entry, and they returned to Jordan."

Earlier Sunday, Walid Attalah, a spokesman for the "Welcome to Palestine" campaign in Jordan told AFP that the two buses had been prevented from crossing by the Jordanians.

"Two buses carrying 100 activists were not allowed to leave the Jordanian side" of Allenby Bridge Crossing, also known as King Hussein Bridge between the West Bank and Jordan, Attalah said.

Atallah said the "activists left the buses and started to demonstrate near the checkpoint. One of them fainted and was taken to hospital."

As part of the campaign, aimed at highlighting Israel's control over entry to the occupied territories, the activists sought to provide Palestinian students with a tonne of toys, pens, notebooks and other supplies.

"This is our fourth initiative to enter Palestine," Zemor told a news conference before leaving for the crossing, around 35 kilometres (20 miles) southeast of Amman.

"We have around 100 participants, from France, Belgium, Spain, Switzerland, the United States, aged between 10 and 50. All kinds of people, from all backgrounds and religions. They are all motivated to visit Palestine."

Five school buses drove the activists to the crossing as they chanted "Free Palestine!" and sang Palestinian traditional songs. Some wore T-shirts with "I have a dream" printed on them, while others flashed V-for-victory sign.

The campaigners tried to enter the occupied West Bank through Israel in July 2011 and April 2012, but the authorities at Tel Aviv's Ben Gurion airport denied them entry.

The activists, who had planned to stay for a week in the West Bank, say their mission comes at the invitation of Bethlehem governor Abdel-Fatah Hamayel.

Some activists had been sceptical in advance they would be allowed in.

"I tried in July last year and April, but the Israelis put me in prison for one day. I expect they will reject us again just like they did before. It is ridiculous and a shame," American volunteer Michael Rabb told AFP.

German Finance Minister rejects more time for Greek reforms again

BERLIN (Reuters) - Germany's finance minister reaffirmed on Saturday his opposition to giving Greece more time to carry out promised reforms, a day after Greek Prime Minister Antonis Samaras pleaded in Berlin for "more air" to implement the painful measures.

"More time generally means more money and that very soon means a new (bailout) program," Wolfgang Schaeuble told the Tagesspiegel on Sunday newspaper, according to advance excerpts of an interview.

"That would not be the right way to solve the fundamental problems of the euro zone," added Schaeuble, noting that the current bailout program agreed at the end of 2011 totaling 130 billion euros is meant to run for three years.

"If after just half a year that were no longer to be sufficient, that would not be a confidence-building measure."

On Friday, Samaras told German Chancellor Angela Merkel during a one-day visit to Berlin that Greece needed more time, not more cash, to meet the terms of its bailout programme.

Merkel said she wanted Greece to stay in the euro zone but gave no sign of agreeing to his request.

She also said the euro zone must await a report from the 'troika' of international lenders due in October before taking further decisions on Greece - a view reiterated on Saturday by French President Francois Hollande after his talks in Paris with Samaras.

Germany, Europe's biggest economy, has grown increasingly impatient with Greece's failure to carry out reforms demanded in return for loans to stave off bankruptcy.

But a leading member of Merkel's centre-right Christian Democrats, Armin Laschet, said the question of whether Greece stayed in the euro zone carried wider implications beyond the purely economic assessment being prepared by the troika.

"(An exit) could lead to instability in a NATO member state. Russia is standing ready with billions to help Greece in such a scenario... Much more is at stake here than just the question of whether Greece meets the criteria (of its bailout)," he told Reuters in comments authorised for publication on Saturday.

Laschet criticized politicians within Germany's centre-right coalition, including Economy Minister Philipp Roesler, leader of the pro-market Free Democrats, for publicly raising the possibility of Greece leaving the euro zone.

Urban Legends and Truths on Health and Fitness - Health, Beauty ...

Imagine the frustration if you were performing some exercise for years and discovered you were doing it wrong. Or, think about the person who incorrectly performs some type of exercise, and that person becomes injured due to incorrect information and execution. These are very real considerations and they happen to people all the time. That is why it?s so important to make sure you get your information from experts and credible sources. Your health and soundness of body depends on it because the world is rampant with bad information in so many areas. The world of health and fitness are no different so here?s some solid information for you.

Information on back pain products and yoga is abundant online. The marketing for yoga states that practicing these poses will help with back pain. This is only true for some cases, making this statement misleading and dangerous. Doing yoga can be helpful if your back pain is caused by muscle problems. Yoga can help you stretch and strengthen the muscles in your back. Your back pain will go away as you do that. Yoga won?t help, however, if your back pain is due to bone problems like an issue with a spinal disc. You can actually make the problem worse by doing yoga in this kind of situation. You are sorely mistaken if you believe that exercise has to be hard to make a difference. Even if your workout isn?t a hard one you will still feel some benefits in a day or two usually. It can be dangerous after years of inactivity to hit the gym hard right away. So it can be dangerous if you have been relatively sedentary and decide to get in shape. There is no reason to rush. In fact, see a doctor and get a quick check-up, then just go slow and you will have a better experience.

Sometime you can hear people grumbling that their body cannot get used to exercising at some specific time of day, or they are too tired at the end of the day. If you know what your natural circadian cycle is you would be able to reset it. Adjustments will come you just have to give it some time. The point is that once you break through your resistance and exercise after work, your body will have more energy to actually do it.

It is important that you be able to identify fact and fiction where your health is concerned. It?s in your best interest to do the research. Before you go out half cocked and exert the energy and time necessary for a change in lifestyle you need to know that you are doing it right. This way you get the optimal benefit to your exercise program.

Furthermore, you can also add variety of gym equipment like Bench Press in your fitness training routine, it can help boosts your physical health and fitness much easier as well as to ensure a much healthy workout in a rapid way. As long as you keep learning, you will naturally move into other areas. What matters most is that you exercise at least a few times every week.

Weight Lifting is a very crucial physical fitness and Frank Porras will help you about this stuff. He will also help you lose weight and get you powerful, lean, and feeling great by using a Stair Stepper.

Reporting Of Dietary Intake Methods In Obesity Trials - Len Saunders

    From Medical Xpress?..

To evaluate the quality of dietary intake methods and reporting, Tracy Burrows, Ph.D., of the University of Newcastle in Australia, and colleagues conducted a systematic review of 31 studies, including 23 reporting energy intake, 20 reporting macronutrient intakes, and 10 reporting food intake outcomes.

The researchers found the overall quality of dietary intake methods reporting to be rated as poor in 52 percent of studies, with only three studies rated as excellent. In these studies the most commonly used methods of assessing diet included the food diary (13), 24-hour recall (five), food frequency questionnaire (four), and dietary questionnaire (four). Food frequency questionnaires were rated as being of higher quality than food diaries or 24-hour recall.

Apple v. Samsung jury finds Apple's patents valid, awards it more than $1.05 billion in damages

The federal court jury in the patent infringement lawsuit between Apple and Samsung has presented its verdict after deliberating for just for 21 hours and 37 minutes following the three week trial. This particular case started with Apple's lawsuit last April and now the jury's decision is that Samsung did infringe on Apple's bounceback patent with all 21 of its products in question. For the patent on pinch-and-zoom, the jury ruled all but three of the devices listed infringed, and more damningly, found that Samsung executives either knew or should have known their products infringed on the listed patents. The jury has also found against Samsung when it comes to Apple's contours on the back of the iPhone and its home screen GUI. The Galaxy Tab however was found not to have infringed upon Apple's iPad design patents. The bad news for Samsung continues, as the jury decided that not only did it willfully infringe on five of the seven Apple patents, but it also upheld their validity.

The amount of the damages against Samsung is in: $1,051,855,000.00. That's less than half of the $2.5 billion it was seeking, but still more than enough to put an exclamation point on this victory for the team from Cupertino.

The jury also ruled that Apple did not infringe upon Samsung's patents with the iPhone 3G and 3GS, and has awarded it zero dollars in damages. We'll have more information for you as it become available.

Promising new drug target discovered for treatment and prevention of heart failure

ScienceDaily (Aug. 25, 2012) ? A promising new drug target for the treatment and prevention of heart failure has been discovered by researchers at Mount Sinai School of Medicine in New York, NY, US.

The study was presented at the ESC Congress 2012 by principal investigator Professor Roger J. Hajjar, MD.

According to the US Centers for Disease Control and Prevention, about 5.8 million Americans suffer from heart failure and 670,000 new cases are diagnosed each year. One in five people with heart failure die within one year of diagnosis. Heart failure is most often treated with aggressive medical and device therapy, but has no cure. The most common symptoms of heart failure are shortness of breath, feeling tired, and swelling in the ankles, feet, legs, and sometimes the abdomen.

In this study presented at the ESC Congress 2012, researchers identified a new drug target that may treat and/or prevent heart failure. The team evaluated failing human and pig hearts and discovered that SUMO1 (small ubiquitin-like modifier), a small protein that regulates the activity of key transporter genes, was decreased in failing hearts. When the researchers injected SUMO1 into these hearts via gene therapy, cardiac function was significantly improved.

"This indicates that SUMO1 may play a critical role in the pathogenesis of heart failure," said Professor Hajjar, who is research director of Mount Sinai's Wiener Family Cardiovascular Research Laboratories.

Led by Professor Hajjar, the team has been evaluating the transporter gene SERCA2a in patients with severe heart failure as part of the CUPID (Calcium Up-regulation by Percutaneous administration of gene therapy In cardiac Disease) trial. When delivered via an adeno-associated virus vector -- an inactive virus that acts as a medication transporter -- into cardiac cells, SERCA2a demonstrated improvement or stabilisation with minimal side effects. But Professor Hajjar said: "We found that while injection with SERCA2a restored cardiac function, over time the new SERCA2a became dysfunctional. This indicated that something else upstream from SERCA2a was causing the dysfunction in the heart."

Dr Changwon Kho, PhD, and Dr Ah Young Lee, PhD, two experts in the study of cardiac proteins at Mount Sinai School of Medicine, identified SUMO1 as the regulator of SERCA2a, showing that it enhanced its function and improved its stability and enzyme activity. When Professor Hajjar and his team studied human and animal models, they found that when SUMO1 was decreased, SERCA2a became dysfunctional in human hearts, showing that SUMO1 plays a protective role. When the team injected SUMO1 as a gene therapy, they found that it protected SERCA2a from oxidative stresses that are prevalent in heart failure.

"Our experiments over the last four years beginning with the discovery of SUMO1 as an interacting protein of SERCA2a have shown that it plays a critical role in the development of heart failure," said Professor Hajjar. "In fact, SUMO1 may be a therapeutic target at the earliest signs of development and may be beneficial in preventing its progression, a much needed advance for the millions suffering from this disease."

Dr Lisa Tilemann extended the experiments performed in mice and rats in a preclinical model of heart failure in porcine models.

Professor Hajjar said: "We have now clearly shown that SUMO1 gene delivery can enhance cardiac function and stabilize the deteriorations of left ventricular volumes in large animals with severe heart failure. We have also shown that delivering SUMO1 and SERCA2a concomitantly can have synergistic benefits on overall function in heart failure."

Led by Professor Hajjar, the Mount Sinai team discovered the landmark potential of SERCA2a in 1999 and since then has been pursuing its potential as a treatment delivered via gene therapy. The next stages in the research include testing a novel gene therapy construct which will combine both SUMO1 and SERCA2a within one gene therapy vector that will enable the investigators to express both genes simultaneously. Similar to their efforts in the CUPID trial they will explore the delivery of SERCA2a and SUMO1 via gene therapy. Additionally, the research team has developed a cellular test to screen for compounds that may increase the interaction of SERCA2a with SUMO1, evaluating SUMO1 as an adjunctive therapy to SERCA2a.

Professor Hajjar concluded: "While this study re-affirms the importance of SERCA2a as a target in heart failure, our discovery of the critical role that SUMO1 plays in improving SERCA2a function in heart failure will hopefully lead to novel treatment strategies for patients with heart failure."
